The quantitative determination of a urinary melatonin metabolite by HPLC in the clinical laboratory has been carried out using Diapak P solid-phase extraction cartridges. The samples were separated on a Eurospher 100-5-C18 column with ultraviolet detection at 223 nm. Eluent composition was acetonitrile-water-isopropyl alcohol (30:70:0.1) (v/v/v), flow rate was 1000 ?L/min, pressure was 100 bar, the procedure was performed at room temperature. Simplicity, reproducibility and sufficient sensitivity of this method combined with a possibility of its implementation on standard chromatography equipment (an isocratic high-pressure pump and a UV detector) makes it suitable for clinical applications.
